Human precision-cut cystic duct and gallbladder slices: a novel method for studying cholangiopathies

چکیده
Background and aims Precision-cut tissue slices (PCTS) are widely used as an ex vivo culture technique to study pathogeneses of diseases drug activities in organs vitro . A novel application the PCTS model may be field translational research into cholangiopathies such biliary atresia. Therefore, aim this was apply precision-cut slice human bile duct gallbladder tissue. Methods Cystic derived from patients undergoing a cholecystectomy were collected, preserved for preparation cystic (PCCDS) (PCGS). The PCCDS PCGS prepared using mechanical slicer subsequently incubated 24 48 h respectively William's Medium E (WME) medium. Viability assessed based on ATP/protein content morphology [hematoxylin eosin (H&E) staining]. Results It shown that viability, by morphology, ( n = 8) could maintained over incubation period respectively. increased significantly 0.58 ± 0.13 pmol/µg at 0 2.4 0.29 after P .0003). similar significant increase 0.94 0.22 3.7 0.41 .0005) 4.2 0.47 .0002) observed PCGS. Double Cystic Duct in a Septated Gallbladder
Double cystic duct in a single gallbladder is one of the least common variances encountered in the biliary system. This article presents a 54-year-old man who had a septated gallbladder with 2 separate cystic ducts. With intraoperative cholangiogram, he had successful laparoscopic cholecystectomy without any ductal injuries or complications.
متن کاملGallbladder drainage and cystic duct stenting
Acute cholecystitis (AC) is one of the commonest presentations to the surgical ward in developed countries. Clinical features, blood analysis and imaging usually lead to an early diagnosis and prompt initiation of antibiotic and supportive medical therapy. Although the clinical course is often self-limiting, complications related to gallbladder perforation, fistulae formation and disseminated s...
متن کاملAgenesis of the gallbladder and cystic duct.
Agenesis of the gallbladder and cystic duct is a rare anomaly that is usually asymptomatic. The patient may present symptoms characteristic of cholelithiasis. Its surgical confirmation requires careful dissection of the common bile duct and intraoperative cholangiography or ultrasonography to be performed, to exclude the possibility of an ectopic gallbladder. The authors describe two cases of t...
متن کاملBiaxial distension of precision-cut lung slices.
The mechanical forces acting on lung parenchyma during (mechanical) ventilation and its (patho)physiological consequences are currently under intense scrutiny. Several in vivo and cell culture models have been developed to study the pulmonary responses to mechanical stretch. While providing extremely useful information, these models do also suffer from limitations in being either too complex fo...
متن کاملA Novel Method for Human Astigmatism Formulation and Measurement
Astigmatism’s defect has not yet been formulated in Physics and medical physics books theoretically. In this paper, we are trying to formulate the astigmatism’s defect. The results of formulation show that “the degree of astigmatism is negative reverse the distance that vision’s condition of astigmatism’s chart lines change conditions in dimness angle from dimness condition at infinite distance...
